Product description: IkappaBalpha is the major IkappaB protein member of IkappaB family that function to inhibit the NF-kappaB transcription factor. IkappaBs have an N-terminal regulatory domain, followed by six or more ankyrin repeats and a PEST domain near their C terminus. IkappaBalpha inhibits NF-kappaB by masking the nuclear localization signals (NLS) of NF-kappaB proteins and keeping them in an inactive state in the cytoplasm. In addition, IkappaBalpha blocks the ability of NF-kappaB transcription factors to bind to DNA, which is required for NF-kappaB's proper functioning. Upon stimulation by the proinflammatory cytokine tumor necrosis factor (TNF), signaling pathways lead to activation of the subunit of the IkappaB kinase (IKK) complex, which then phosphorylates IkappaB proteins on two N-terminal serine residues. The IKK is an unusual kinase in that it contains two related kinases, IKKalpha and IKKbeta, and a regulatory subunit, NEMO (NF-kappaB essential modifier). The primary role of NF-kappaB is to maintain normal cellular functions that range from cell-to-cell communication to cell motility, cell cycle progression, and cell lineage development. Inhibits the activity of dimeric NF-kappa-B/REL complexes by trapping REL dimers in the cytoplasm through masking of their nuclear localization signals. On cellular stimulation by immune and proinflammatory responses, becomes phosphorylated promoting ubiquitination and degradation, enabling the dimeric RELA to tranlocate to the nucleus and activate transcription.
